How to avoid hidden dangers of cryopreservation tubes? What are the methods?

In laboratories, samples such as cells and tissues often need to be packaged in cryovials and stored in ultra-low temperature or liquid nitrogen environments. However, when taking it out, the cryovial tube sometimes bursts suddenly, which may not only cause the loss of precious samples, but also cause serious injuries to the experimenters. Therefore, understanding the causes and countermeasures of #cryotube bursts is particularly critical for experimental safety.



Part.1/The root cause of cryopreservation tube burst


In experiments, cells usually need to be stored at -130°C or lower for long periods of time. The temperature of the gaseous layer of the liquid nitrogen tank is between -140°C and -180°C. You can choose to store the cells in the gaseous layer or directly immerse them in liquid nitrogen.



#The main reason why cryopreservation tubes burst is that liquid nitrogen accidentally remains in the tubes. After taking out the cryovial, the liquid nitrogen vaporizes rapidly, and 1 volume of liquid nitrogen can expand into approximately 646 volumes of nitrogen. If the cryopreservation tube cannot be vented in time, the internal pressure will increase suddenly, and when the tube body cannot withstand it, it will burst.



Aijin Bio-freezing tubes are made of imported medical-grade polypropylene (PP) material, which has excellent low-temperature resistance and sealing design. It can be used stably in the temperature range of -196°C to 121°C, greatly reducing the risk of tube explosion and improving safety of use.




Part.2/ The difference between internal rotation and external rotation cryopreservation tubes


Tightness is a key factor when selecting a cryovial type. Structurally, when liquid nitrogen vaporizes, it is easier to discharge in the internal rotation cryogenic tube, and the sealing performance is generally better than that of the external rotation tube, so it is not easy to cause explosion in the liquid nitrogen environment.



However, the bottom of the inner screw cap may come into contact with the sample, posing a certain risk of contamination; while the outer screw cap can avoid contact with the sample due to its threaded shell, improving operational cleanliness.





Part.3/How to reduce the risk of bursting of externally rotated cryopreservation tubes


If you only have external rotation cryovials, the following operations can effectively reduce the probability of tube explosion:



Take out the cryopreservation tube from the liquid nitrogen and place it horizontally or upside down, wait for tens of seconds for the liquid nitrogen to be fully discharged, and then open the tube cap;

Increase the volume of freezing solution appropriately (for example, add 1.8 mL of freezing solution to a 2 mL cryopreservation tube), keep a small amount of gas space, avoid excessive pressure, and tighten the tube cap as much as possible when freezing cells;

③The use of cryopreservation tube sleeves can isolate the tube body from direct contact with liquid nitrogen to a certain extent, thereby preventing liquid nitrogen from penetrating into the tube;

Choose liquid nitrogen gas layer storage to reduce the possibility of liquid nitrogen infiltration, but only a small amount of liquid nitrogen can be stored in liquid nitrogen, and liquid nitrogen needs to be added frequently.
