The purification column consists of a centrifuge tube, a preparation tube, a pressure ring, and a silica gel membrane. The silica gel membrane in the middle plays a key role. The silica gel membrane can selectively adsorb nucleic acids, and this process is reversible.

Among many spin columns, the silica gel membrane is the main nucleic acid adsorption medium. It is actually a layer of glass fiber with a large number of modified silanol groups (Si-OH) on the surface. It will be destroyed in a high-salt environment and thus adsorbed on the spin column, while other impurities such as proteins will be precipitated by centrifugation.
