In the centrifugal column method of nucleic acid extraction, the purification column is the core component that determines the quality of extraction, and its interior is usually filled with a silica gel membrane matrix with high specific adsorption capacity. The silica gel membrane can specifically bind nucleic acid molecules through hydrophobic interaction, hydrogen bonding and the like in a high-salt buffer environment, and at the same time repel impurities such as proteins, polysaccharides, lipids and the like; when a low-salt eluent is subsequently added and driven by centrifugal force, the nucleic acid molecules will be separated from the silica gel membrane and dissolved in the eluent to achieve efficient purification.
